【摘要】:1.1Thisguidedescribesproceduresforobtaininglaboratorydataconcerningtheadverseeffectsofatestmaterialaddedtoseawater,butnottofood,onthemarinecopepodAmphiascustenuiremis,duringcontinuousexposuresofindividuals,fromimmediatelyafterbirth,untilafterthebeginningofreproductionusinga200956;Lrenewalmicroplate-culturingtechnique.Thefollowingdataarecheckedandrecordedduringthetestperiod:stage-specificsurvival,numberofdaysittakesfordevelopmentfromafirststagenaupliustoareproductivelymaturecopepod,genderratios,numberofdaysforafemaletoextrudefirstandsubsequentbroods,numberofdaysbetweenfirst(andsubsequent)broodextrusion(s)andhatchingoffirst-generationnauplii,numberofhatchedandsurvivingnauplii,numberofunhatchedornecroticeggsandabortedunhatchingeggsacs,andthetotalnumberoffemalesabletoproduceviableoffspringovertheentirematingperiod.Thismicroplate-basedfulllife-cycletoxicitytesthasadurationofapproximately17daysfortoxicantsthatdonotdelaydevelopment.Theseproceduresprobablywillbeusefulforconductinglife-cycletoxicitytestswithotherspeciesofcopepods,althoughmodificationsmightbenecessary.1.2Theseproceduresareapplicabletomostchemicals,eitherindividually,orinformulations,commercialproducts,orknownmixtures,thatcanbemeasuredaccuratelyatthenecessaryconcentrationinwater.Withappropriatemodificationstheseprocedurescanbeusedtoconducttestsontemperature,dissolvedoxygen,andpHandonsuchmaterialsasaqueouseffluents(seealsoGuideE1192),sedimentporewaters,andsurfacewaters.Renewalmicroplatetestsmightnotbeapplicabletomaterialsthathaveahighoxygendemand,arehighlyvolatile,arerapidlytransformed(biologicallyorchemically)inaqueoussolutions,orareremovedfromtestsolutionsinsubstantialquantitiesbythetestchambersororganismsduringthetest.Iftheconcentrationofdissolvedoxygenfallsbelow50%ofsaturation,ortheconcentrationoftestmaterialinthetestsolutiondecreasesbymorethan20%betweenrenewals,itmightbedesirabletorenewthesolutionsmoreoften.1.3Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatoryrequirementspriortouse.
